1685-Glioma Metabolism Drives Neuronal Activity

Dela

Recent scientific findings reveal that high-grade gliomas accumulate massive amounts of the metabolic intermediate guanidinoacetate (GAA) due to imbalanced enzyme activities in the creatine synthesis pathway. Rather than producing creatine, tumor cells heavily secrete GAA into the surrounding brain tissue, mirroring conditions seen in inborn errors of metabolism like GAMT deficiency. Researchers discovered that this secreted GAA acts as an agonist for GABA A receptors, triggering depolarizing currents in peritumoral neurons that display dysregulated chloride homeostasis. Consequently, this metabolic pathway drives neuronal hyperexcitability and establishes critical neuron-tumor interactions that accelerate cancer growth. Experiments demonstrate that genetic depletion or knockout of AGAT—the enzyme responsible for GAA production—significantly reduces electrochemical signaling and extends survival in orthotopic xenograft models. Furthermore, artificial neuronal stimulation can rescue the decreased tumor aggressiveness caused by GAA loss, highlighting a direct link between cancer metabolism and neuroscience.
