Transdermal Therapy Targets Acidity of Tumor Microenvironment to Enhance Immunotherapy Outcomes with Dr. Ryan Beal Dyve Biosciences TRANSCRIPT

Dela

Dr. Ryan Beal, Founder and CEO of Dyve Biosciences, has developed a transdermal delivery approach that uses the skin to deliver agents into circulation, where they selectively neutralize the acidic tumor microenvironment. By correcting acidity, the immune system is reactivated to better recognize and attack cancer cells, as acidity serves as a natural immune checkpoint that allows tumors to evade immune surveillance. Unlike previous oral delivery approaches that cause gastrointestinal side effects, this non-invasive lotion-based technology delivers the same pharmaceutical safety and efficacy.

Ryan explains, "What led me there is that tumor acidity has driven cancer biology for decades. The science has settled. What was missing was a way to actually fix it in patients and do it safely, broadly, and at scale. And that's what Dyve built. In 2026, the goal is to bring it into humans with trials at Moffitt Cancer Center."

"The core at Dyve was developing the delivery system. It could bring molecules through the skin. And one of the approaches that we got quite excited about early on was delivering buffering agents through the skin. The natural application was in oncology and going after the acidity of the tumor microenvironment."

"So it's a topical. In many ways it will look and feel like a lotion, and it's applied to the skin but delivered systemically. So we're essentially turning the skin into a delivery portal for these drugs. And that's important, especially important in the case of tumor acidity.  Because we and others have shown that while in animals you can deliver these therapies orally, it's a big delivery challenge for humans to take these agents orally. So you needed another way to circumvent those problems, and the skin gave us a really convenient path to do that."
